In the acute form, occurring within 100 d after transplant, GVHD is a signifi cant cause of transplant-related mortality; as a chronic disease, occurring sometimes indefi nitely from 100 d after transplant, it generates long-term morbidity (1). In murine models, inactivation of recipient class I MHC prevents the initiation of acute GVHD (2) and selective class I MHC or costimulatory antigen knockout implicates both recipient and donor APCs in the full expression of chronic GVHD (3, 4). More specifi c analysis of cutaneous GVHD has revealed the potent function of recipient Langerhans cells (LC) and the attenuation of acute GVHD by temporal separation of donor engraftment in the LC and T cell compartments (5). The potential persistence of recipient LC after HSCT

In conventional descriptions, there is a division of labor between DC, which are dedicated to the induction and control of adaptive immunity ( 1, 2 ), and macrophages, which are occupied with the cl...

In this issue of JEM, Wu et al. (https://doi.org/10.1084/jem.20171341) use genetic barcoding of macaque hematopoietic stem cells to demonstrate that, after transplantation, HSCs are very asymmetrically distributed and uncover a thymus-independent pathway for mature T cell production in the bone marrow.
متن کاملTransient apoptosis inhibition in donor stem cells improves hematopoietic stem cell transplantation
During hematopoietic stem cell transplantation, a substantial number of donor cells are lost because of apoptotic cell death. Transplantation-associated apoptosis is mediated mainly by the proapoptotic BCL-2 family proteins BIM and BMF, and their proapoptotic function is conserved between mouse and human stem and progenitor cells. Permanent inhibition of apoptosis in donor cells caused by the l...
متن کاملA Review of Procedures Involved in Human Umbilical Cord Blood Banking and Transplantation
Cord blood hematopoietic stem cells are widely used as an alternative source for hematopoietic stem cells transplant. Increasing rate of patients who need hematopoietic stem cells transplant and many advantages of cord blood in comparison to bone marrow hematopoietic stem cells, have promoted banking of cord blood units. Cord blood banking requires accurate steps in donor selection, cord blood ...
متن کاملETV2/ER71 regulates hematopoietic regeneration by promoting hematopoietic stem cell proliferation
Recent studies have established that hematopoietic stem cells (HSCs) are quiescent in homeostatic conditions but undergo extensive cell cycle and expansion upon bone marrow (BM) transplantation or hematopoietic injury. The molecular basis for HSC activation and expansion is not completely understood. In this study, we found that key developmentally critical genes controlling hematopoietic stem ...
